Abstract:
Abstract: Objective To study the radiosensitization effect of mefloquine on glioma cells and to explore its mechanism. Methods The glioma U251 cells at the exponential growth period were sampled and their survival rate after mefloquine action was studied by CCK-8 assay cell clone formation experiments were used to evaluate the radiosensitization effect of mefloquine on U251 cells flow cytometry was used to detect the active oxygen level and apoptosis rate of U251 cells after mefloquine combined with X-ray, and the death mechanism of U251 cells under the combined effect was explored. Results Mefloquine had a good radiosensitization effect on glioma cells, and its maximum radiosensitization ratio to U251 cells was 1.64. After X-ray combined with mefloquine acted on U251 cells, the level of cell apoptosis increased, and the level of reactive oxygen species increased. The active oxygen inhibitor glutathione can inhibit U251 cell apoptosis induced by X-ray combined with mefloquine. Conclusion Mefloquine has a radiosensitizing effect on U251 cells, and it is probably through increasing the level of cellular reactive oxygen species and inducing apoptosisthat mefloquinecan lead to increased radiosensitivity.
