Abstract:
Abstract: Objective To discuss the variations of the expression of matrix metalloproteinase-1 (MMP-1) and tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) in osteoblasts with the increasing loading time of fluid shear stress (FSS). Methods MC3T3-E1 cells were subjected to FSS (12 dyn/cm2) using a parallel plate flow system for 0, 15, 30, 45, 60 min. The expression level of MMP-1 and its inhibitor TIMP-1 were detected by Western blotting. Results FSS of 12 dyn /cm2 was applied to the MC3T3-E1 cells cultured in vitro. With the increase of loading time, the expression of MMP-1 was up-regulated, while the expression of TIMP-1 was down-regulated. Forty-five minutes after loading, the expression of MMP-1 was the maximum and that of TIMP-1 reached the minimum. Conclusion Loading 12 dyn /cm2 FSS can promote the expression of MMP-1, and down-regulates the expression of TIMP-1 in MC3T3-E1 cells. The optimal loading time of FSS is 45minutes.
